**INTENDED USE AND TEST PRINCIPLE**
This Human Corin ELISA Kit is designed for research purposes only and is not intended for diagnostic or therapeutic use. The kit utilizes a sandwich ELISA method to quantitatively measure human Corin levels in various biological samples. A standard curve is generated using known concentrations of Corin, allowing for accurate calculation of unknown sample concentrations based on optical density (OD) readings at 450 nm.
**SAMPLE COLLECTION AND STORAGE**
- **Serum**: Collect using a serum separator tube. Allow samples to clot for 2 hours at room temperature or overnight at 4°C before centrifugation at 2000×g for 20 minutes. Assay immediately or store at -20°C. Avoid repeated freeze-thaw cycles.
- **Plasma**: Use heparin as an anticoagulant. Centrifuge within 30 minutes of collection at 2000×g for 30 minutes at 2–8°C. Store at -20°C. Avoid multiple freeze-thaw cycles.
- **Cell culture supernatants, tissue homogenates, and other body fluids**: Centrifuge to remove particulates and assay immediately or store at -20°C. Ensure no hemolysis or granulation occurs.
**MATERIALS REQUIRED BUT NOT SUPPLIED**
1. 37°C incubator
2. Microplate reader capable of measuring absorbance at 450 nm
3. Precision pipettes, disposable tips, and absorbent paper
4. Distilled or deionized water
**REAGENTS PROVIDED**
All reagents should be stored at 2–8°C. Check expiration date on the label.

**PRECAUTIONS**
- Do not mix reagents from different kits.
- Do not thaw samples or reagents in a water bath.
- Do not use beyond the expiration date.
- Only use deionized or distilled water for dilution.
- Keep microtiter plates in sealed bags until use.
- Use fresh pipette tips for each transfer to avoid contamination.
- Wear disposable gloves during the entire procedure.
- Waste must be inactivated for at least 30 minutes before disposal.
- Substrate solutions are sensitive; discard if discolored.
- Substrate B contains 20% acetone—keep away from heat or flame.
- Allow all reagents to reach room temperature (20–25°C) before use.
**REAGENT PREPARATION AND STORAGE**
- **Wash Solution (1X)**: Dilute 1 volume of 20X Wash Solution with 19 volumes of deionized or distilled water. Store at 2–8°C for up to 1 month.
**ASSAY PROCEDURE**
1. Prepare all reagents before starting.
2. Add 100 µL of standards, samples, and blanks to the microtiter plate. Cover with adhesive strips and incubate for 60 minutes at 37°C.
3. Wash the plate 4 times using either manual or automated washing methods.
4. Add 50 µL of Chromogen A and 50 µL of Chromogen B to each well. Incubate for 15 minutes at 37°C, protected from light.
5. Add 50 µL of Stop Solution to each well. The color should change from blue to yellow. Gently tap the plate if the color change is uneven.
6. Measure OD at 450 nm within 15 minutes using a microplate reader.
**CALCULATION OF RESULTS**
- Plot the average OD values of the standards against their concentrations to create a standard curve.
- Subtract the blank OD value from all measurements before interpretation.
- Use graph paper or software to determine sample concentrations.
- Intra-assay and inter-assay CV% are less than 15%.
- Assay range: 1.56 ng/mL to 50 ng/mL.
- Sensitivity: <1.0 ng/mL.
- Cross-reactivity: No significant cross-reaction with other proteins.
- Storage: 2–8°C (for frequent use); 6 months at -20°C.
**NOTES**
- If sample values exceed the highest standard, dilute with Sample Diluent and repeat.
